Neither!
Would you please do me the great honour of reviewing my thesis?
If you are asking that question, reviewing the thesis will be no small
task!

Neither.
Would you do me the honor of reviewing my thesis?

Any version using "honor" sounds patronizing to me. There's no honor
involved. Oh, there might be if you are asking some renowned figure
in the field who is so famous and respected that it honors you that he
is willing to glance at your work, but - in most cases - you are just
asking a favor of some ordinary person. If the person is ordinary,
using "honor" that way sounds almost sarcastic.

It does sound a bit smarmy but I think the OP could get away with
"Would you do me the honour". It does sound very archaic, though.
To be very humble what about: "Would you be so kind as to review my
thesis?"

Yes, but it's quite normal in Chinese and Japanese, which is why the
question: the poster is from Hong Kong, it seems.

I think you had better say: 'I'd be honoured if you reviewed my
thesis.'
'Would you do me the honour of reviewing my thesis' might be slightly
off because it suggests that you intend for the person addressed to
participate in your opinion that he is doing you an honour which he
needn't -- he may just be doing you a kindness -- and you ought not to
expect him to.
